Why Your Leaky Gut Won’t Heal (And What Your Genes Have to Do With It)

The Histamine Connection: HNMT, DAO, and MAO-B

Three of these genes form what we call the histamine trio. HNMT, DAO, and MAO-B all play roles in breaking down histamine, that inflammatory compound your body releases during allergic reactions. Here’s what most people don’t know: when your body can’t properly break down histamine, it doesn’t just cause hives and sneezing—it creates a vicious cycle with your gut barrier.

Research shows that people with histamine intolerance consistently have elevated zonulin levels, the protein that regulates those gap junctions in your gut lining. When you have variants in HNMT, DAO, or MAO-B, your body can’t clear histamine efficiently. It builds up in your system alongside increased gut permeability, creating a self-perpetuating cycle: the more your gut leaks, the more inflammation you have; the more inflammation, the harder it is for your body to maintain proper barrier function.
